چکیده
Culture fluids from five muminesarcomas were chemotac tic for syngeneic penitoneal macnophages in vitro. Penito neal macmophages from mice infected with Mycobacterium bovis, strain Bacillus Ca!mette-Guérin,were more respon sive to the chemotactic factor in tumor cultures than were normal macnophages. Penitoneal gran ulocytes, however, did not significantly respond to this factor. The level of chemotactic activity in tumor cultures paralleled cell growth for all five tumors; maximal levels occurred during log growth. Culture medium alone on fluids from proliferating spleen cell cultures stimulated with mitogens did not have detectable chemotactic activity. Chromatography of the tu momculture fluids resulted in a single peak of chemotactic activity in the i5,000-moleculam weight mangeon Sephadex G-iOO and at about 7.5 mmho/cm specific conductance on diethylaminoethyl cellulose. By both biological and physi cochemical characteristics, the chemotactic activity in tu mon culture fluids was different from mouse lymphocyte derived chemotactic factor.

Listeria monocytogenes (LM) in admixture with B-16 mel anoma suppresses local tumor development in syngeneic CS7BL/6 mice. In vitro, LM-immune pemitoneal and splenic cells are cytotoxic to B-16. Induction of cell-mediated im munity to LM antigens are required fan the killing effect, since effectom cells from LM-―immune― athymic nude mice are unable to kill tumor cells in vitro. Further, eli...
متن کاملIsolation, Molecular Identification And Genomic Pattern of Mycobacterium Bovis Isolates Collected From Tuberculin-Positive Cattle in Infected Farms of Shiraz
Background: Mycobacterium bovis is the main cause of tuberculosis in cattle. At the global scale and also in Iran, the most frequent currently-in-use method in the detection of infected cattle is tuberculination. Objective: The present study was aimed to improve our genomic knowledge of Mycobacterium bovis population structure in cattle farms of Shiraz. Materials and Methods: Fifty pathologic...
متن کاملAntigen-reactivity pattern of T-cell hybridomas from Mycobacterium bovis BCG-infected mice.
Lymph node cells from mice infected with live Mycobacterium bovis BCG were fused with BW5147 cells after short-term culturing in vitro. Both mycobacterium- and self-reactive T-cell hybridomas were identified. Some T-cell hybridoma clones displayed dual reactivity to self and to self plus mycobacterial antigen but did so to a different degree, indicating that infection with mycobacteria stimulat...
متن کاملIn vivo and in vitro administration of interleukin 2-containing preparation reverses T-cell unresponsiveness in Mycobacterium bovis BCG-infected mice.
Mice infected with high doses of Mycobacterium bovis BCG (3 X 10(7)) showed a marked impairment of delayed-type hypersensitivity to PPD in vivo, and their splenic T cells failed to proliferate when cultured in vitro with concanavalin A or PPD. However, this state of unresponsiveness could be reversed both in vitro and in vivo by the administration of an interleukin 2 (IL-2)-containing preparati...
متن کاملLeukemia L1210 Therapy Trials with Antileukemia Serum and Bacillus Calmette-Guérin1
Further experiments on the therapy of leukemia L1210 with rabbit antiserum prepared against whole leukemia cells have confirmed the mild effectiveness of this agent, which gave no long-term survivors. In contrast, therapy with Mycobacterium bovis (strain Bacillus Calmette-Guérin) gave some long-term survivors, when used under optimal conditions of route and of other still undefined factors, wh...
